Is it required to perform both an optical and SEM analysis?

No. Either optical or SEM analysis can be performed without the other, however what makes the most sense is to perform an optical analysis first. Once it is seen that there are specific particles of interest, you can review those findings and decide if you want to have an SEM analysis performs on those particles.
